Abstract

The spontaneous frequency of pigment-deficient mutants was 5,7×10-5 in haploid protoplast cultures of Nicotiana plumbaginifolia. This frequency could be increased to 8×10-3 by60Co irradiation. In the same system an isoleucine (ILE401), a uracil (URA401) and a leucine (LEU403) auxotroph were recovered by individual testing. Pigment deficiency (5 clones tested) and the auxotrophic traits were shown to be recessive by fusion-complementation. The suitability of haploid Nicotiana plumbaginifolia is discussed for studies on mutation induction in cultured cells.
